NMN Research Explained: What Have Human Clinical Trials Actually Found?
Our most detailed guide: what randomised human trials have reported about NMN and NAD+, physical performance, insulin sensitivity and safety — and where the evidence stops.
NMN has generated enormous interest in longevity and NAD+ research.
But there's a problem.
Laboratory research, animal studies and human clinical trials are frequently discussed as though they provide the same level of evidence.
They don't.
Let's look specifically at what researchers have found in humans.
Human evidence involving NMN and NAD+
One of the clearest areas of research concerns NAD+.
A randomised placebo-controlled trial involving 30 healthy adults investigated 250mg NMN daily for 12 weeks.
Researchers reported significantly increased whole-blood NAD+ concentrations in the NMN group.

A larger trial involving 80 middle-aged adults investigated placebo, 300mg NMN, 600mg NMN and 900mg NMN.
Blood NAD concentrations increased across the NMN groups during the trial.

This provides human evidence that oral NMN can influence NAD+ biology.

The 80-person trial also measured physical performance using a six-minute walking test.
Researchers reported differences in walking-distance changes between groups during the 60-day study.
These results are scientifically interesting.
However, the trial was relatively small and short, and the findings should not be turned into a guarantee that NMN improves exercise performance.
NMN and insulin sensitivity research
One frequently discussed clinical trial involved postmenopausal women with prediabetes who were overweight or obese.
Researchers investigated NMN for ten weeks and reported changes in insulin-stimulated glucose disposal and skeletal-muscle insulin signalling.

This generated considerable research interest.
But one study doesn't settle a scientific question.
What happens when multiple metabolic studies are combined?
Systematic reviews and meta-analyses allow researchers to examine results across multiple trials.
Later analyses have not consistently demonstrated significant improvements across fasting glucose, insulin, HbA1c and lipid-related outcomes.
This is why scientific findings need to be interpreted as a body of evidence rather than selecting one promising study.
NMN and safety
Human trials have generally reported favourable short-term tolerability.
The 250mg, 12-week trial reported no obvious adverse effects within the study's monitored parameters.

Studies involving higher doses over shorter periods have also contributed to the available safety data.
But short-term tolerability is not equivalent to established long-term safety.
Longer studies are needed.

Those conclusions go substantially beyond current human evidence.

NMN research continues to develop.
Larger trials, longer follow-up periods and better-standardised outcomes should provide a clearer picture of whether changes in NAD+ translate into meaningful long-term effects.
