NMN Benefits: What Does the Science Actually Say?

A measured look at the human research behind NMN — NAD+, ageing, physical performance, metabolic and cardiovascular outcomes, and what remains unproven.

Search for NMN benefits online and you'll encounter some extraordinary claims.

Energy. Longevity. Metabolism. Exercise performance. Healthy ageing.

But what has actually been demonstrated in humans?

The answer is more complicated — and more interesting — than many supplement advertisements suggest.

1. NMN and NAD+

The strongest reason for scientific interest in NMN is its relationship with NAD+.

NMN is a precursor involved in NAD+ biosynthesis.

NAD+ itself participates in cellular metabolism and numerous biochemical reactions.

Human studies have reported that oral NMN supplementation can influence NAD+-related measurements.

A placebo-controlled study investigating 250mg NMN daily for 12 weeks reported significantly increased whole-blood NAD+ concentrations.

This provides evidence that orally consumed NMN can affect the biochemical pathway researchers intended to target.

However, increased NAD+ should not automatically be interpreted as proof of every other benefit attributed to NMN online.

NMN and NAD+: What's the Connection?

2. NMN and ageing research

NMN has become closely associated with longevity research.

Scientists are interested in how NAD+ metabolism relates to ageing and whether manipulating NAD+ pathways could have meaningful biological effects.

Preclinical research has produced interesting findings.

Human evidence is less conclusive.

A 2025 Nature Metabolism review concluded that although preclinical research surrounding NAD+ precursors is promising, clinical evidence remains limited.

NMN is therefore better described as an active area of ageing research rather than a proven intervention for reversing ageing.

3. NMN and physical performance research

Researchers have investigated outcomes including walking performance and muscle-related measurements.

One randomised 60-day trial involving 80 healthy middle-aged adults included a six-minute walking test among its outcomes while comparing placebo with several NMN doses.

The results contributed to scientific interest in NMN and physical performance.

Individual trials, however, should not be interpreted as proof that NMN will improve athletic performance for everyone.

4. NMN and metabolic research

Researchers have also investigated NMN in relation to glucose metabolism, insulin sensitivity and other metabolic markers.

Some individual trials have reported interesting results in particular populations.

However, when multiple studies are considered together, the picture becomes less clear.

Systematic reviews and meta-analyses have not consistently demonstrated significant improvements across common metabolic outcomes.

That illustrates why it is important to consider the totality of evidence rather than a single promising study.

5. NMN and cardiovascular research

Cardiovascular-related measurements have also been investigated.

For example, a 12-week placebo-controlled study involving healthy middle-aged adults investigated 250mg NMN per day and measured arterial stiffness alongside NAD+-related metabolites.

Some measurements moved in potentially interesting directions, but differences were not consistently statistically significant.

This should not be interpreted as evidence that NMN prevents cardiovascular disease.

What hasn't been proven?

Current human evidence does not establish NMN as a treatment for:

  • ageing
  • diabetes
  • cardiovascular disease
  • neurological disease
  • sleep disorders
  • other medical conditions

NMN research is developing rapidly, but the evidence needs to be interpreted carefully.
